Human Anti-HIV-1 gp41 Recombinant Antibody (clone 2F5)

Recombinant Human Antibody (2F5) is capable of binding to MPER of HIV-1 gp41, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-HIV-1 gp41 mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-HIV-1 gp41 mAb and CL of human light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

A positive bar indicates an affinity increase and a negative bar indicates an affinity decrease. The bars are color-coded for each ligand Mab. For context, the MPER residues comprising the 2F5 and 4E10 epitopes are shown and the residues 669–673 scanned by the alanine substitutions are as indicated. Figure 6 Neutralizing activity of MPER-specific IgA1 on primary and laboratory HIV-1 strains. 4E10 and 2F5 were used as positive controls. Dose-dependent neutralization was performed within a concentration range of 0.156–10 mg/ml.
